SENP7在多种生物学过程中发挥着重要作用。例如,SENP7可以感知氧化应激,维持CD8+ T细胞的代谢状态和抗肿瘤功能[1]。SENP7缺陷的CD8+ T细胞表现出糖酵解和氧化磷酸化下降,导致细胞增殖能力减弱和抗肿瘤功能受损。SENP7还可以通过去SUMO化PTEN蛋白,促进其降解,从而防止PTEN依赖性代谢缺陷。此外,SENP7还可以感知细胞内ROS水平,调节肌动蛋白组装和肌肉萎缩[2]。SENP7缺陷会导致Fli-I蛋白SUMO化水平升高,降低Safb1在染色质上的定位,从而影响肌球蛋白重链(MyHC-IId)基因的表达,导致肌小节组织和肌肉收缩功能受损。SENP7还可以通过去SUMO化cGAS蛋白,促进其激活,进而激活STING信号通路,参与先天免疫反应[3]。SENP7还可以通过调节NLRP3炎症小体和NF-κB信号通路,抑制细胞的焦亡[5]。SENP7在神经发生过程中也发挥着重要作用,参与神经元分化[6]。SENP7缺陷会导致神经元分化受阻,影响神经系统的发育。SENP7基因的突变会导致多种严重的先天性疾病,包括致死性多发性关节挛缩症、早期呼吸衰竭和中性粒细胞减少症等[4][7][8][9]。
