在缺氧的肿瘤微环境中,ESM1的表达水平升高,通过PKM2依赖的Warburg效应促进卵巢癌细胞的脂肪酸合成和血管生成拟态。具体来说,缺氧通过转录激活HIF-1α诱导ESM1表达上调,而ESM1作为PKM2和UBA2相互作用的关键介质,促进PKM2的SUMO化修饰和二聚体形成,进而促进Warburg效应和PKM2的核转位,最终导致STAT3的磷酸化,从而促进卵巢癌细胞的糖酵解和血管生成拟态[2]。

在食管癌中,ESM1的表达水平显著升高,与患者的预后不良相关。ESM1敲低可以抑制食管癌细胞的增殖、迁移和侵袭。此外,ESM1的基因多态性与食管癌的易感性相关[3]。

在头颈鳞状细胞癌中,ESM1的表达水平显著升高,与血管生成相关。ESM1的共表达基因中富集了多个转录因子结合位点,包括SMAD3、SMAD4、SOX3、SOX4、HIF2A和AP-1等。进一步分析发现,AP-1是ESM1表达的重要调控因子[4]。

FOXO1可以刺激内皮细胞中尖端细胞富集基因的表达,包括ESM1和ANGPT2。FOXO1通过VEGF介导的尖端细胞定义在原代培养的内皮细胞中发挥作用[5]。

在黑色素瘤中,ESM1的表达水平显著升高,与患者的预后不良相关。ESM1敲低可以抑制黑色素瘤细胞的增殖、迁移和侵袭。此外,ESM1的表达与免疫相关细胞和基因的增多,以及肿瘤进展通路如Notch和Wnt的激活相关[6]。

在乳头状甲状腺癌中,ESM1的表达水平显著升高,与患者的预后不良相关。ESM1敲低可以抑制甲状腺癌细胞的增殖、迁移和侵袭[7]。

在泛癌分析中,ESM1在12种癌症类型中表达上调,包括膀胱癌、乳腺癌、结直肠癌、胆囊癌、食管癌、头颈鳞状细胞癌、肾透明细胞癌、肾嫌色细胞癌、肝细胞癌、胃腺癌、胸腺癌和子宫内膜癌。ESM1的表达与多种癌症患者的总生存期和疾病无进展生存期相关[8]。

在宫颈癌中,ESM1的表达水平显著升高,与患者的预后不良相关。ESM1敲低可以抑制宫颈癌细胞的增殖、迁移和侵袭,并通过抑制PI3K-Akt通路的激活和EMT的进展来抑制宫颈癌的发生和发展[9]。

综上所述,ESM1在多种肿瘤的发生和发展中发挥重要作用,可以作为一种新的肿瘤标志物和治疗靶点。未来需要进一步研究ESM1在肿瘤发生和发展中的具体作用机制,为肿瘤的治疗和预防提供新的思路和策略。
