Jdp2,也称为Jun dimerization protein 2,是一种重要的转录因子,属于AP-1家族。它通过与其他c-Jun蛋白形成二聚体,抑制AP-1复合物的转录活性,从而在细胞分化、增殖、细胞凋亡、氧化应激反应和肿瘤发生等方面发挥重要作用。

Jdp2在多种生物学过程中发挥作用。首先,Jdp2在AHR-NRF2基因电池中发挥关键作用。AHR和NRF2是两种重要的转录因子,可以调节一系列基因的表达,编码解毒I期和II期酶。Jdp2与AHR-AHR核转位因子(ARNT)和NRF2-小肌肉腱膜纤维肉瘤蛋白(sMAF)形成转录复合物,然后被招募到各自的顺式元件,如二噁英反应元件和抗氧化反应元件,从而激活AHR基因的转录。Jdp2还在AHR-NRF2复合物从二噁英反应元件转移到AHR启动子中的抗氧化反应元件中发挥作用,从而激活AHR启动子[1,2]。其次,Jdp2具有组蛋白伴侣活性,可以抑制组蛋白乙酰转移酶的活性,从而抑制组蛋白乙酰化和核小体组装。Jdp2还抑制多梳抑制复合物1和2(PRC-1和PRC-2)募集到p16(Ink4a)/Arf基因座,从而抑制p16(Ink4a)/Arf基因的表达,进而抑制细胞衰老[3]。此外,Jdp2还参与谷胱甘肽代谢的调控。在基因毒性应激下,β-catenin与ATM磷酸化-JDP2和PRMT5形成复合物,招募WDR5/MLL甲基转移酶复合物,并在GSH代谢相关基因的启动子上诱导H3K4甲基化,从而促进谷胱甘肽的合成和红氧化平衡的重建[4]。

Jdp2在多种疾病中发挥重要作用。在肝细胞癌(HCC)中,Jdp2的表达水平与肿瘤大小和分期呈负相关,高表达的Jdp2与更好的生存率相关,提示Jdp2可能是一种肿瘤抑制因子[5]。Jdp2的表达受p53的负调控,而Jdp2又可以抑制p53的表达,从而形成一个负反馈回路。Jdp2的表达也受ATF4的直接调控,Jdp2可以抑制ATF4介导的基因表达,从而降低细胞对TNF相关凋亡诱导配体(TRAIL)的敏感性,从而抑制细胞凋亡[6,7]。Jdp2还可以抑制其同源基因ATF3的表达,从而影响细胞对多种应激和生长刺激的适应性反应[8]。

Jdp2是一种重要的转录因子,在多种生物学过程中发挥作用,包括解毒、细胞衰老、氧化应激反应和肿瘤发生等。Jdp2在多种疾病中发挥重要作用,包括肝细胞癌、动脉粥样硬化、糖尿病心肌病、结直肠癌和Wilms瘤等。此外,Jdp2还具有独立的染色质调控功能,影响基因表达和干细胞的多能性维持。Jdp2的研究有助于深入理解转录因子的生物学功能和疾病发生机制,为疾病的治疗和预防提供新的思路和策略。
