Bdh2(3-羟基丁酸脱氢酶2)是一种短链脱氢酶/还原酶家族成员,参与多种生物学和病理学过程。在哺乳动物中,Bdh2催化哺乳动物铁载体的生物合成中的一个速率限制步骤,在铁稳态、能量代谢和细胞凋亡中发挥关键作用。Bdh2的表达和功能在不同组织和细胞类型中有所不同,其调控机制和生物学功能尚不完全清楚。

在胃癌中,Bdh2的表达显著下调,低表达与患者的不良生存率相关。Bdh2过表达可显著诱导细胞凋亡和自噬,抑制胃癌细胞的生长[1]。机制上,Bdh2促进Keap1与Nrf2的相互作用,增加Nrf2的泛素化水平,抑制ARE的活性,进而增加活性氧(ROS)的积累,抑制PI3K/Akt/mTOR信号通路的磷酸化水平,从而抑制胃癌的生长。

在类风湿关节炎(RA)中,Bdh2的表达下调,HBEGF+成纤维细胞减少。HBEGF+成纤维细胞在RA缓解中发挥作用,通过EGF信号通路与滑膜巨噬细胞相互作用,促进组织修复[3]。

在酵母中,BDH2基因的过表达可降低啤酒中二乙酰的含量,减少不良的黄油风味[4]。BDH2基因的过表达还可以提高酵母对香草醛的耐受性,改善发酵效率[8]。此外,BDH1和BDH2基因的过表达可以降低葡萄酒中二乙酰的含量,提高葡萄酒质量[9]。

在胚胎干细胞(ESC)中,Bdh2的缺失导致端胚分化潜能升高,上调端胚相关基因的表达。Bdh2缺失还抑制了多种DNA甲基转移酶(DNMTs)的表达,提示Bdh2可能对维持DNA甲基化至关重要[5]。

在系统性红斑狼疮(SLE)中,Bdh2的表达下调,细胞内铁水平升高,DNA羟甲基化水平升高,甲基化水平降低。Bdh2下调通过增加细胞内铁,导致CD4+ T细胞中DNA羟甲基化和低甲基化,进而导致免疫相关基因的过表达[6]。
