STARD3,全称为StAR相关脂质转运结构域包含蛋白3,是一种在细胞内发挥胆固醇转运和代谢作用的蛋白质。该基因编码的蛋白质属于STARD蛋白家族,包含一个与类固醇生成急性调节蛋白(StAR)相关的脂质转运结构域,负责将胆固醇从内质网转运到内体,在维持细胞胆固醇稳态中发挥重要作用。

在乳腺癌中,STARD3基因与人类表皮生长因子受体2(HER2)基因共扩增,其表达水平与HER2阳性乳腺癌肿瘤和细胞系显著相关。研究表明,STARD3的高表达与HER2阳性乳腺癌患者的总生存期、无病生存期和疾病无转移生存期较差相关。此外,STARD3的低表达与雌激素受体阳性(ER+)乳腺癌患者的总生存期较差相关,提示STARD3可能对HER2阳性乳腺癌具有重要的诊断和预后价值[1,3,4,6]。

除了在乳腺癌中的作用,STARD3在其他癌症类型中也有报道。研究发现,STARD3基因位于人类染色体17q12的PPP1R1B-STARD3-ERBB2-GRB7扩增子上,与胃癌的发生发展密切相关。STARD3基因在该扩增子上与PPP1R1B、ERBB2和GRB7等基因共扩增,共同促进胃癌的发生[5,9]。

除了在癌症中的作用,STARD3还在其他生物学过程中发挥作用。例如,研究发现,胆固醇过量与慢性阻塞性肺疾病(COPD)和气道上皮细胞驱动的炎症相关。STARD3通过调节STARD3-MFN2信号通路,影响胆固醇的重新分布、转运和积累,以及线粒体功能和动力学,从而影响气道上皮细胞对炎症的反应[2]。此外,STARD3还参与视网膜色素上皮细胞中胆固醇的代谢和转运,其表达水平的降低可能与年龄相关性黄斑变性(AMD)的发生发展相关[7]。

STARD3还参与脂肪生成过程。研究发现,STARD3在3T3-L1前脂肪细胞的脂肪生成过程中表达显著增加,同时伴随着线粒体活性氧(ROS)的产生增加。STARD3敲低可以抑制3T3-L1前脂肪细胞的脂肪生成,并降低线粒体ROS水平,提示STARD3在脂肪生成过程中通过促进胆固醇向线粒体的再分配,增加线粒体ROS的产生而发挥作用[8]。

综上所述,STARD3是一种重要的蛋白质,参与胆固醇转运和代谢,在维持细胞胆固醇稳态中发挥重要作用。STARD3在乳腺癌、胃癌、慢性阻塞性肺疾病和脂肪生成等生物学过程中发挥作用,其表达水平与多种疾病的发病机制和预后相关。因此,STARD3可能成为一种新的生物标志物和治疗靶点,为相关疾病的治疗和预防提供新的思路和策略。
