在其他研究中,Chen等人[3]在台湾地区对245例孤立或合并肌张力障碍患者进行了IMPDH2基因分析,未发现致病性变异,提示IMPDH2在肌张力障碍中可能不起主要作用。然而,Zech等人[4]对764例肌张力障碍患者进行全外显子测序,发现IMPDH2基因中的6个变异与肌张力障碍相关,支持IMPDH2在神经发育障碍中的潜在作用。Lin等人[5]对688例中国肌张力障碍患者进行了研究,发现IMPDH2基因中的4个罕见变异与肌张力障碍相关,进一步证实了IMPDH2在肌张力障碍中的重要作用。

此外,Sugimoto等人[6]发现IMPDH2基因的表达在Epstein-Barr病毒(EBV)感染的原发性B细胞中上调,导致核仁肥大和细胞增殖。IMPDH2的抑制剂Mycophenolic acid(MPA)和Mycophenolate mofetil(MMF)可以抑制EBV诱导的B细胞增殖,表明IMPDH2在EBV相关疾病中的作用。Gao等人[7]的研究表明,IMPDH2在双打击淋巴瘤(DHL)中表达上调,通过激活PI3K/AKT/mTOR信号通路促进肿瘤发展。Huang等人[8]的研究发现,Wnt/β-catenin信号通路激活IMPDH2介导的嘌呤代谢,通过抑制caspase依赖性凋亡,促进结直肠癌对奥沙利铂的耐药性。

Liu等人[9]的研究通过整合大脑衍生的分子定量性状位点(mRNA表达和蛋白质丰度定量性状位点)和大规模全基因组关联研究(GWAS)的遗传发现,识别了25个精神疾病的潜在药物靶点。其中,IMPDH2基因被确定为抑郁症的潜在药物靶点,为精神疾病的治疗提供了新的思路。最后,Xu等人[10]的研究发现,IMPDH2基因的缺失可以抑制破骨细胞生成,减轻卵巢切除引起的骨质疏松症,提示IMPDH2可能在骨代谢中发挥重要作用。

综上所述,IMPDH2基因在多种疾病中具有重要作用,包括神经发育障碍、肿瘤、EBV相关疾病、精神疾病和骨质疏松症等。IMPDH2基因的突变、表达异常和信号通路调控与这些疾病的发病机制密切相关。因此,深入研究IMPDH2基因的功能和作用机制,有助于揭示这些疾病的发病机制,为疾病的治疗和预防提供新的思路和策略。
