STK10(Serine/Threonine Kinase 10),也称为淋巴细胞定向激酶(LOK),是一种在STE20家族中具有丝氨酸/苏氨酸激酶活性的独特成员。STE20家族的激酶在细胞的生长、凋亡和形态发生等多种细胞功能中发挥重要作用。STK10在人类和鼠类中表达主要局限于淋巴样细胞,而在人类中,其在多种组织中也存在转录本,尽管在造血细胞中表达水平最高[9]。STK10的基因编码区位于人类染色体5q35.1,在鼠类位于染色体11A4,在鼠类中,STK10基因的3'非翻译区存在多态性CA重复序列,这有助于进一步定位STK10基因在染色体上的位置[2]。

STK10基因的突变还与获得性纯红细胞再生障碍(PRCA)的发病机制有关。研究发现,STK10基因的敲低可以抑制红系分化和促进K562细胞的凋亡,这可能是通过抑制核糖体生物合成和降低核糖体水平来实现的[4]。此外,STK10基因的突变还与抗凋亡功能有关。研究发现,STK10基因的一个错义突变(R634H)可以显著降低STK10抑制NF-κB活性和增强地塞米松诱导的凋亡的能力[6]。

STK10在急性髓系白血病(AML)中的表达与不良预后相关。研究发现,STK10在AML中过表达,并且STK10表达水平高的患者总体生存率较差。此外,STK10还与免疫细胞浸润和免疫检查点有关,如HLA-E、CD274和GAL-9[7]。

在果蝇中,STK10的直系同源基因Slik被发现可以防止JNK信号通路介导的细胞凋亡,从而维持组织稳态。研究发现,Slik的缺失可以激活JNK信号通路,导致细胞凋亡和组织稳态受损。此外,STK10的敲低也可以激活人癌细胞中的JNK信号通路,而表达Slik可以消除这种激活[8]。

综上所述,STK10在细胞粘附、迁移、凋亡和组织稳态等方面发挥重要作用。STK10的突变或缺失可能与多种疾病的发病机制有关,包括哮喘、宫颈癌、前列腺癌、获得性纯红细胞再生障碍和急性髓系白血病等。因此,STK10可能成为这些疾病诊断、预后评估和治疗的潜在靶点。
