PTPRK,即蛋白酪氨酸磷酸酶κ(Protein Tyrosine Phosphatase Receptor Kappa),是一种受体型蛋白酪氨酸磷酸酶,属于蛋白酪氨酸磷酸酶家族。PTPRK通过去除磷酸基团,调节多种信号转导途径,从而影响细胞生长、分化、迁移和侵袭等多种生物学过程。PTPRK在多种细胞和组织中表达,包括免疫系统细胞、上皮细胞和肿瘤细胞等。

PTPRK在T细胞发育中发挥重要作用。研究发现,在LEC大鼠中,PTPRK基因的缺失会导致T细胞发育缺陷,表现为T辅助细胞(Th)的免疫缺陷。具体来说,PTPRK基因的缺失会导致胸腺中T细胞从双阳性(DP)细胞向CD4单阳性(SP)细胞的成熟过程受阻[1,4]。此外,PTPRK的缺失还与早期结直肠癌的发生发展相关。研究发现,早期结直肠癌患者中,PTPRK基因存在与RSPO3基因的融合,这种融合导致PTPRK基因的表达下调,进而影响Wnt信号通路,促进肿瘤细胞的生长和增殖[2]。

PTPRK在多种疾病中发挥重要作用。研究发现,PTPRK的缺失与阿尔茨海默病(AD)的发病风险相关。在一项多人群全基因组关联分析中,PTPRK基因被确定为AD的新的风险基因之一[5]。此外,PTPRK的表达下调与霍奇金淋巴瘤(HL)的发生发展相关。研究发现,EB病毒(EBV)感染可以下调PTPRK的表达,从而促进HL细胞的生长和存活[6]。PTPRK的表达还与妊娠并发症——先兆子痫(PE)的发生风险相关。研究发现,PTPRK基因的某些单核苷酸多态性(SNPs)与PE的发生风险相关[7]。

PTPRK在Wnt信号通路中发挥重要作用。研究发现,PTPRK可以与锌指蛋白RING3(ZNRF3)相互作用,调节Wnt受体蛋白的降解和Wnt信号通路的活性。具体来说,PTPRK可以去除ZNRF3上的磷酸基团,促进ZNRF3的内吞和降解,从而抑制Wnt信号通路。相反,MET激酶可以磷酸化ZNRF3,抑制PTPRK的活性,从而激活Wnt信号通路[8]。
