在卵巢癌、非小细胞肺癌和结直肠癌等多种肿瘤中,CCR8+ Treg细胞在肿瘤组织中显著富集,且高表达与不良预后相关[4,7,8]。CCR8+ Treg细胞表达多种免疫检查点分子,如PD-1配体1和PD-1配体2,以及CCR8趋化因子受体,这些分子在Treg细胞抑制抗肿瘤免疫反应中发挥重要作用[4,8]。此外,CCR8+ Treg细胞还表达IL-1受体2,这提示CCR8+ Treg细胞可能通过IL-1信号通路抑制抗肿瘤免疫反应[8]。

CCR8靶向治疗是一种有潜力的肿瘤免疫治疗策略。抗CCR8抗体能够特异性地清除肿瘤组织中CCR8+ Treg细胞,从而恢复CD4+和CD8+ T细胞的抗肿瘤免疫活性,抑制肿瘤生长[1,2,3]。在结直肠癌中,抗CCR8抗体治疗能够有效地抑制肿瘤生长,并且与PD-1阻断疗法联合使用时,表现出更强的抗肿瘤效果[6]。在卵巢癌中,CCR8+ Treg细胞的清除能够显著抑制肿瘤生长,且未观察到明显的毒性反应[4]。

综上所述,CCR8在肿瘤浸润性Treg细胞中特异性表达,并在肿瘤免疫抑制中发挥重要作用。CCR8靶向治疗能够特异性地清除肿瘤组织中CCR8+ Treg细胞,从而恢复抗肿瘤免疫反应,抑制肿瘤生长。CCR8靶向治疗是一种有潜力的肿瘤免疫治疗策略,值得进一步研究和开发。
