ATP5PF,也称为ATP合酶外周柄亚基F6,是线粒体ATP合酶的一个关键组成部分。ATP合酶是负责合成细胞能量货币——ATP的酶,位于线粒体内膜上。ATP合酶由多个亚基组成,其中ATP5PF是外周柄亚基之一,它与其他亚基协同作用,通过氧化磷酸化过程将质子梯度转换为ATP的合成。ATP5PF的这种功能对于维持细胞能量代谢至关重要,尤其是在能量需求较高的细胞类型中,如心肌细胞、骨骼肌细胞和癌细胞。

ATP5PF的表达和功能异常与多种疾病的发生和发展密切相关。例如,在非小细胞肺癌(NSCLC)中,ATP5PF的表达上调与癌细胞增殖和迁移有关[1]。研究表明,POU3F3蛋白在NSCLC细胞中的核转位被ERK1磷酸化所介导,并激活ATP5PF的转录,进而增强ATP的产生,从而促进NSCLC的进展。

在寻常型天疱疮(PV)中,ATP5PF的表达也显著增加[2]。研究发现,PV患者皮肤样品中的氧化磷酸化(OXPHOS)途径被激活,而ATP5PF等OXPHOS相关蛋白的表达上调。这表明OXPHOS途径可能在天疱疮的发病机制中发挥重要作用。

此外,ATP5PF的表达还与唐氏综合症(DS)相关[3]。研究显示,DS患者羊水细胞的ATP5PF表达水平升高,提示ATP5PF可能与DS相关的发育障碍有关。

在乳腺癌中,ATP5PF的表达也与患者的预后相关[4]。研究发现,ATP5PF等OXPHOS相关基因的表达上调与乳腺癌患者的较差生存率相关。此外,ATP5PF的表达还与乳腺癌肿瘤微环境的变化有关,如TGF-β和mTOR信号通路的激活。

在分娩过程中,ATP5PF的表达上调也与子宫收缩有关[5]。研究发现,SERPINE1基因通过增加ATP的产生来增强子宫收缩力,而SERPINE1与ATP5PF相互作用,共同维持ATP的产生。

在山羊的瘤胃上皮细胞中,ATP5PF的表达也与瘤胃的发育相关[6]。研究发现,外源性β-羟基丁酸酸(BHBA)的补充可以促进瘤胃上皮细胞的生长和代谢,而ATP5PF等OXPHOS相关基因的表达上调可能参与了这一过程。

在低流量低梯度主动脉瓣狭窄(PLF-LG)的心肌中,ATP5PF的表达也上调[7]。研究发现,PLF-LG患者心肌细胞和细胞外基质中的ATP5PF等OXPHOS相关蛋白表达上调,提示OXPHOS途径可能与PLF-LG的发病机制相关。

在哈萨克马肌肉的发育过程中,ATP5PF的表达也受到年龄的影响[8]。研究发现,随着年龄的增长,最长背肌中的ATP5PF表达水平下调,提示ATP5PF可能与肌肉的代谢和功能有关。

在强直性脊柱炎(AS)中,ATP5PF的表达也与神经退行性疾病相关[9]。研究发现,AS患者血液中的ATP5PF等OXPHOS相关基因表达上调,提示OXPHOS途径可能在AS和神经退行性疾病中发挥共同的作用。

综上所述,ATP5PF作为线粒体ATP合酶的关键组成部分,参与调控细胞的能量代谢。ATP5PF的表达和功能异常与多种疾病的发生和发展密切相关,包括非小细胞肺癌、寻常型天疱疮、唐氏综合症、乳腺癌、分娩过程、瘤胃发育、主动脉瓣狭窄和强直性脊柱炎等。ATP5PF的研究有助于深入理解细胞能量代谢的生物学功能和疾病发生机制,为疾病的治疗和预防提供新的思路和策略。
