此外,Fancd2的缺陷与同源重组修复(HR)缺陷相关,这类缺陷的肿瘤细胞对PARP抑制剂敏感。PARP抑制剂通过抑制PARP介导的DNA修复途径,导致HR缺陷的肿瘤细胞发生合成致死效应,从而抑制肿瘤的生长。在卵巢癌中,Fancd2基因的缺失导致肿瘤细胞对Polθ介导的修复途径依赖,这为PARP抑制剂和Polθ抑制剂联合使用治疗卵巢癌提供了理论基础[3]。

Fancd2还与铁死亡(ferroptosis)相关。铁死亡是一种铁依赖性的细胞死亡形式,在多种疾病中发挥作用。在肾小管损伤修复过程中,FANCD2和RAD51等HR相关基因的表达上调,而FANCD2的缺失会导致不完全修复和肾脏纤维化,从而加剧慢性肾脏疾病(CKD)的进展[6]。在心肌缺血再灌注损伤中,FANCD2的表达受ATF3转录因子的调控,ATF3通过增加FANCD2启动子的活性抑制铁死亡,发挥心脏保护作用[7]。
