Cytohesin-1(Cyth1),是一种小分子G蛋白,属于ARF(ADP-ribosylation factor)激活蛋白家族,主要功能是激活ARF蛋白,参与细胞骨架重组和膜转运过程,包括G蛋白偶联受体(GPCR)的内吞作用[8]。Cyth1的表达和功能与多种生物学过程相关,如细胞粘附、迁移、归巢和植入等。

此外,Cyth1还在急性髓性白血病(AML)中发挥重要作用。研究发现,Cyth1在AML患者中显著过表达,与不良预后相关。敲低Cyth1的表达可以显著抑制AML细胞的粘附、迁移、归巢和植入能力,延缓疾病进展,延长动物存活时间。Cyth1抑制剂SecinH3可抑制AML细胞的增殖,促进细胞凋亡,并与BCL2选择性抑制剂ABT-199协同作用,抑制ABT-199耐药的AML细胞的生长[2]。

Cyth1还与心脏生物学相关。研究发现,Cyth1基因在BALB/cByJ小鼠中存在自然缺失,但该缺失并未影响心肌细胞的多倍体水平[3]。

Cyth1在人类造血干细胞和祖细胞(HSPCs)中也发挥重要作用。Cyth1敲低可导致HSPCs的粘附、归巢和植入能力降低,影响其在骨髓中的定位和定居[4]。

此外,研究发现吸烟者的尿液总尼古丁当量(TNEs)与CYTH1基因的甲基化水平相关,提示Cyth1可能参与吸烟相关的病理过程[5]。

在阿尔茨海默病(AD)中,miR-125b-5p作为炎症调节因子,可以调节多个与AD相关的信号通路,其中包括CYTH1基因[6]。

在乳羊奶中,CYTH1基因的SNP与乳酸含量相关,提示CYTH1可能参与乳羊奶的代谢过程[7]。
