NEO1,也称为Neogenin-1,是一种重要的细胞表面受体蛋白。它属于免疫球蛋白超家族成员,与已知的肿瘤抑制基因DCC(Deleted in Colorectal Cancer)密切相关。NEO1蛋白由四个免疫球蛋白样(Ig-like)结构域、六个纤连蛋白III型结构域、一个跨膜结构域和一个细胞内结构域组成。NEO1基因位于人类染色体15q22.3-q23区域,与Bardet-Biedl综合征相关基因位点邻近。NEO1蛋白在胚胎发育过程中表达于神经细胞,并在其他胚胎组织中也有表达,提示其可能参与组织生长调控、细胞识别和迁移等发育过程[6]。

NEO1蛋白在多种生物学过程中发挥重要作用。在神经发育过程中,NEO1作为轴突导向信号分子Netrin-1的受体,参与神经元迁移和轴突导向。研究发现,Netrin-1通过与Neo1和Unc5b受体共同调节Wnt和MAPK信号通路,促进小鼠胚胎干细胞(mESCs)的幼稚多能性维持[3]。此外,NEO1还在造血系统中发挥作用,区分小鼠长期造血干细胞(LT-HSCs)中具有髓系偏向和平衡分化潜力的细胞亚群。NEO1+Hoxb5+ LT-HSCs在年龄增长和骨髓抑制应激后表现出髓系偏向分化,而NEO1-Hoxb5+ LT-HSCs则保持平衡分化能力[2]。

NEO1在肿瘤发生发展中也具有重要作用。研究发现,NEO1在结直肠癌(CRC)组织中表达下调,与患者不良预后相关。NEO1表达下调与炎症相关信号通路、上皮-间质转化(EMT)和血管生成等肿瘤相关过程相关。过表达NEO1可以抑制CRC细胞的增殖、迁移和侵袭,而NEO1表达缺失则表现出相反的效果[1]。此外,NEO1还在其他类型的肿瘤中发挥作用,如头颈部鳞状细胞癌(HNSCC)。研究发现,HNSCC患者中NEO1的转录表达水平与局部疾病控制相关,低表达NEO1的患者具有更高的局部复发风险[7]。

NEO1在非神经系统和炎症反应中也具有重要作用。研究发现,NEO1在肝缺血再灌注损伤(IRI)中发挥关键作用。Neo1基因敲除小鼠和骨髓嵌合动物在肝IRI过程中表现出损伤减轻,血清乳酸脱氢酶、天冬氨酸和丙氨酸氨基转移酶水平降低,炎症因子水平下降,肝组织学评分改善。此外,使用抗Neo1抗体处理小鼠也显著减轻了肝IRI[4]。此外,NEO1还与铁代谢相关。研究发现,肝细胞中的NEO1与血红素结合蛋白Hemojuvelin(HJV)相互作用,参与肝脏铁调素的表达和铁稳态的调节。肝细胞特异性敲除Neo1导致铁调素表达下降和铁超载[5]。
