FOXA3在癌症的发生发展中也有重要作用。在胰腺癌中,FOXA2/3基因的表达上调,提示FOXA3可能参与了胰腺癌的发生发展。在肺腺癌中,FOXA3通过调节胆固醇代谢来补偿低摄入,从而促进肺腺癌的转移。FOXA3通过转录激活Hmgcs1基因,诱导胆固醇生物合成,改变细胞膜成分,进而促进细胞迁移。此外,FOXA3的抑制剂能够抑制肺腺癌患者来源的类器官(PDOs)中的转移基因程序[3][4]。

FOXA3在肝脏再生和细胞重编程中也发挥着重要作用。研究发现,FOXA3、HNF1A和HNF4A三种转录因子可以共同作用,将成纤维细胞直接重编程为具有肝细胞功能的细胞,称为人诱导肝细胞(hiHeps)。这些hiHeps细胞表达肝细胞基因程序,可以体外扩增,并具有成熟肝细胞的功能,如细胞色素P450酶活性和胆汁药物清除能力。将hiHeps细胞移植到小鼠模型中,可以恢复肝脏功能,延长生存时间[6]。

FOXA3在生殖系统中也有重要作用。研究发现,宫内暴露于内分泌干扰化学物混合物(EDCs)会影响成年大鼠睾丸中FOXA3基因和蛋白的表达。FOXA3是Leydig细胞功能的关键调节因子,其表达下调可能导致成年大鼠睾丸中Leydig细胞功能受损,进而影响雄性生殖功能[7]。

FOXA3在结直肠癌中也发挥着重要作用。研究表明,FOXA3是结直肠癌分子亚型的一个关键转录因子。在iCMS3亚型的结直肠癌中,FOXA3的表达上调,并与肿瘤的发生发展密切相关[5]。
