在系统性红斑狼疮中,GAS5基因多态性与SLE风险相关。研究发现,GAS5基因启动子区域的5个碱基对(AGGCA/-)插入/缺失(I/D)多态性(rs145204276)与SLE风险降低相关。此外,GAS5的表达水平在SLE患者中下调,而携带rs145204276 ID/DD基因型的患者表现出GAS5和PTEN表达水平的上调。这些发现表明,GAS5/miR-21/PTEN信号通路参与了SLE的发生,其中GAS5作为miR-21的靶点,miR-21调节PTEN的表达[2]。

在尿路上皮细胞癌中,GAS5基因多态性与UCC发展相关。研究发现,女性UCC患者中,携带rs145204276 GAS5 Ins/Del或Del/Del基因型的患者发生大肿瘤状态的风险比携带rs145204276野生型(Ins/Ins)基因型的患者高3.037倍。此外,GAS5的表达水平在女性膀胱尿路上皮癌患者中较低,与肿瘤大小呈负相关。因此,GAS5基因多态性可能成为预测女性UCC患者临床状态的指标[3]。

在器官纤维化和动脉粥样硬化中,GAS5也发挥着重要作用。研究发现,GAS5在器官纤维化中发挥作用,并且与miRNA和lncRNA的调节有关。此外,GAS5在动脉粥样硬化中也发挥着重要作用,通过调节巨噬细胞的炎症反应来影响动脉粥样硬化的发生发展[4,5]。

GAS5作为一种lncRNA,在多种人类癌症中发挥着重要的肿瘤抑制作用。GAS5的表达水平在肿瘤组织中降低,导致肿瘤恶性程度增加、预后不良和耐药性增强。GAS5通过多种分子机制发挥肿瘤抑制作用,包括调节细胞增殖、侵袭、迁移和凋亡等。此外,GAS5基因多态性也与癌症易感性相关。因此,GAS5的研究对于深入理解肿瘤发生发展机制和寻找新的治疗靶点具有重要意义[6,7,8,9,10]。
