Sprr3，即小富含脯氨酸蛋白3，是富含脯氨酸蛋白家族的一员，属于角化包膜前体蛋白。该家族成员在表皮角质形成细胞的终末分化过程中发挥重要作用，Sprr3主要表达于口腔和食管上皮组织中，与角质形成细胞的终末分化密切相关。Sprr3基因具有独特的结构特征，包括基因内重复导致的脯氨酸富集序列，一个替代的非编码外显子，以及在启动子区域存在一个200bp的嘧啶富集区。这些结构特征与Sprr3基因的表达调控密切相关。

Sprr3基因的表达受到多种调控因子的控制。研究发现，Sprr3基因启动子区域存在多个调控元件，包括高亲和力Ets结合位点、AP-1结合位点和ATF/CRE结合位点。这些调控元件分别由ESE-1、Jun/Fos家族转录因子和Jun/Fos、ATF因子识别并结合，共同调控Sprr3基因的表达。此外，Sprr3基因启动子区域还发现两个新的调控元件，一个与GAAP-1结合，抑制Sprr3转录，另一个与钙离子结合，激活Sprr3启动子活性。

Sprr3在多种疾病中发挥重要作用。研究发现，Sprr3在胰腺癌中表达上调，与患者不良预后相关。Sprr3的过表达可以促进胰腺癌细胞的无锚定生长能力、生长速度和侵袭能力，而Sprr3的敲低则表现出相反的结果。进一步研究发现，Sprr3通过NF-κB信号通路发挥其促癌作用，过表达SPRR3可以促进NF-κB下游基因的转录，增加p65蛋白在细胞核中的积累。因此，靶向Sprr3可能成为治疗胰腺癌的有效策略[1]。

Sprr3还在过敏性疾病中发挥作用。研究发现，Sprr3在过敏性气道炎症条件下表达上调。通过基因敲低实验发现，Sprr3的敲低可以显著抑制哮喘小鼠肺组织中嗜酸性粒细胞的浸润和2型细胞因子的表达。进一步研究发现，Sprr3通过IL-33/ILC2轴和PI3K/AKT/NF-κB信号通路促进过敏性气道炎症。因此，Sprr3可能成为哮喘治疗的潜在靶点[2]。

Sprr3还与头颈鳞状细胞癌（HNSCC）的发生发展相关。研究发现，Sprr3在HNSCC中表达上调，与CD8+ T细胞和巨噬细胞的浸润水平相关。Sprr3的表达水平与HNSCC患者的总生存期相关，提示Sprr3可能作为HNSCC的潜在诊断和预后生物标志物[6]。

此外，Sprr3在肺癌、结直肠癌和乳腺癌等多种恶性肿瘤中发挥重要作用。研究发现，Sprr3在肺癌、结直肠癌和乳腺癌中表达上调，与患者的预后不良相关。Sprr3通过PI3K/AKT信号通路促进肺癌、结直肠癌和乳腺癌细胞的增殖、迁移和侵袭。因此，Sprr3可能成为这些恶性肿瘤的潜在治疗靶点[3,4,5,7,8,9]。

综上所述，Sprr3作为一种重要的角化包膜前体蛋白，在多种生物学过程中发挥重要作用，包括表皮角质形成细胞的终末分化、过敏性气道炎症、头颈鳞状细胞癌、肺癌、结直肠癌和乳腺癌等。Sprr3的表达受到多种调控因子的控制，其功能与NF-κB信号通路、PI3K/AKT信号通路和IL-33/ILC2轴等密切相关。因此，Sprr3可能成为多种疾病的潜在治疗靶点，为疾病的治疗和预防提供新的思路和策略。
