RASSF7,也称为HRC1,是人类11号染色体上一个重要的基因,属于Ras相关结构域家族(RASSF)的N末端成员。RASSF家族是一组在肿瘤抑制中发挥关键作用的基因,其成员在多种生物学过程中发挥重要作用,包括细胞周期控制、细胞凋亡、微管动力学和细胞增殖分化等[1,2]。RASSF7作为N末端RASSF家族的一员,在肿瘤发生发展中扮演着重要的角色。

RASSF7在多种恶性肿瘤中表现出异常表达。研究表明,RASSF7在肺癌、神经母细胞瘤、甲状腺肿瘤、肝细胞癌、乳腺癌和胃癌等多种恶性肿瘤中表达水平升高[3]。RASSF7的表达水平与肿瘤的TNM分期呈正相关,提示其在肿瘤进展中发挥重要作用。此外,RASSF7的表达还与肿瘤的预后相关。在神经母细胞瘤中,RASSF7的表达水平与MYCN基因扩增和INRG分期M相关,提示其可能作为神经母细胞瘤诊断和预后评估的潜在分子标志物[4]。

RASSF7在肿瘤发生发展中的作用机制复杂。研究发现,RASSF7可以调节多种信号通路,包括Aurora B、MKK4、MKK7、JNK、YAP、MEK和ERK等,从而影响细胞周期、细胞凋亡和细胞增殖分化等过程[3]。例如,RASSF7可以通过调节Aurora B激酶的活性,影响有丝分裂的进行;通过激活MEK1/2-ERK1/2信号通路,促进肝细胞癌细胞的增殖;通过与c-Myc蛋白相互作用,抑制其功能,从而抑制肿瘤的发生发展[5,10]。

除了在肿瘤发生发展中的作用,RASSF7还与胎盘基因表达相关。研究表明,孕期暴露于交通相关空气污染(TRAP)会影响胎盘基因表达,其中RASSF7的表达水平与胎儿性别和孕期暴露于二氧化氮(NO2)水平存在交互作用[6]。此外,RASSF7的表达水平还与三阴性乳腺癌(TNBC)的转移相关。研究发现,TNBC组织中RASSF7的表达水平升高,且与肿瘤转移相关基因的表达水平呈正相关[7]。

RASSF7在细胞凋亡中也发挥着重要作用。研究发现,在退行性间盘病变(IDD)中,RASSF7的表达水平降低,且过表达RASSF7可以抑制细胞凋亡的发生[8]。这提示RASSF7可能通过调节细胞凋亡参与IDD的发生发展。

RASSF7在细胞有丝分裂中也发挥着重要作用。研究发现,RASSF7是细胞有丝分裂所必需的,其功能缺失会导致细胞无法形成有丝分裂纺锤体,进而导致细胞周期阻滞、细胞凋亡和组织结构破坏[9]。这提示RASSF7可能通过调节细胞有丝分裂参与肿瘤的发生发展。

综上所述,RASSF7在肿瘤发生发展中发挥着重要作用,其表达水平与肿瘤的分期、预后和转移相关。RASSF7通过调节多种信号通路和生物学过程,影响细胞周期、细胞凋亡、细胞增殖分化和细胞有丝分裂等过程,从而参与肿瘤的发生发展。此外,RASSF7还与胎盘基因表达相关,提示其在胎儿发育中可能发挥重要作用。深入研究RASSF7的功能和作用机制,有助于揭示肿瘤发生发展的分子机制,为肿瘤的诊断、预后评估和靶向治疗提供新的思路和策略。
