SPAG6在癌症的发生和发展中也发挥重要作用。研究发现,SPAG6基因在多种癌症组织中表达异常,包括急性髓系白血病、肺癌和结直肠癌等。SPAG6基因的甲基化水平与癌症的发生和发展密切相关,DNA甲基转移酶DNMT3b介导的SPAG6启动子高甲基化会导致SPAG6基因表达下调,从而促进癌症的发生和发展[4,7,10]。此外,SPAG6基因还与癌症细胞的干性相关,SPAG6表达水平的升高会抑制癌症细胞干性的标记基因的表达,从而抑制癌症的发生和发展[7]。
