近年来,UBE2K在多种疾病中的作用逐渐受到关注。在抑郁症的发生发展中,circ-UBE2K被发现能够与HNRNPU形成复合物,上调UBE2K的表达,进而导致异常的微胶质细胞激活和神经炎症,促进抑郁症的发生和发展[1]。在阿尔茨海默病中,circRIMS2被发现能够通过激活GluN2B的泛素化来介导突触和记忆障碍。UBE2K是circRIMS2下游的靶基因,其表达上调能够导致突触功能障碍[2]。在肝细胞癌中,UBE2K被发现能够与HIF-1α形成功能轴,促进肝细胞癌的恶性进展[3,4]。此外,UBE2K还在胰腺导管腺癌和胶质瘤中发挥重要作用,促进肿瘤细胞的增殖和干性[5,6]。

此外,UBE2K还参与了神经发生过程中组蛋白H3的降解。在人类胚胎干细胞中,UBE2K表达上调能够促进组蛋白H3的泛素化降解,降低H3K9me3的抑制性标记,从而影响神经发生过程[7]。在胶质瘤细胞中,UBE2K表达上调能够抑制自噬相关的细胞凋亡,从而促进胶质瘤细胞的生长[8]。

在多发性骨髓瘤中,Ixazomib被发现能够通过靶向UBE2K来抑制骨髓瘤细胞的增殖。Ixazomib能够降低UBE2K的表达,进而影响细胞周期、细胞凋亡和活性氧的产生[9]。

综上所述,UBE2K是一种重要的泛素结合酶,在多种疾病中发挥重要作用。UBE2K的异常表达和功能改变可能导致细胞增殖、凋亡、炎症和神经功能障碍等病理过程。因此,UBE2K可能成为治疗多种疾病的新靶点,为疾病的治疗和预防提供新的思路和策略[1-10]。
